1.0 Purpose
1.1 To describe a procedure for operation and calibration of UV Cabinet.
2.0 Scope
2.1 This procedure is applicable for operation and calibration of UV Cabinet located at Instrumentation
Room in Quality Control laboratory at ABC Pvt. Ltd.
3.0 Responsibility
Officer QC/ Executive QC Preparation of SOP and follow the procedure as per SOP
Assistant Manager Reviewing and Training of SOP

4.0 Accountability
4.1 Manager Quality Control is accountable for implementation and compliance with SOP.
5.0 Procedure
5.1 Operation
5.1.1 Ensure that the instrument is free from dust.
5.1.2 Switch on the mains.
5.1.3 Switch ON the instrument indicator light will be ON. After that off the switch.
5.1.4 Open the cabinet and place the TLC plate inside and close the cabinet.
5.1.5 Three switches are provided on the top right corner of the instrument.
Switch 1: For UV-Light at long wavelength
Switch 2: For UV-Light at short wavelength
Switch 3: For Visible
5.1.6 Select the required UV-Light and switch ON.
5.1.7 Observe the spot from the TLC plate.
5.1.8 After that switch off the instrument.
5.2 Calibration (Frequency: after 6 Months)
5.2.1 Ultra-Violet lamps used for viewing of Thin Layer Chromatography can be calibrated.
5.2.2 Prepare a 0.04% w/v solution of sodium salicylate in alcohol and apply a 5 micro litre spot
on a plate coated with silica gel GF-254 for lamps emitting emission band with maximum intensity at about 254 nm.
5.2.3 Place the TLC plate under the radiation so that the radiation falls perpendicularly on the plate and examine the spot.
5.2.4 The lamp should be capable of revealing a standard spot of a diameter of about 5 mm of the plate.
5.2.5 Prepare a 0.02% w/v solution of sodium salicylate in alcohol and apply a 5 micro litre spot on
a plate coated with silica gel GF-254 for lamps emitting emission band with maximum intensity at about 365 nm.
5.2.6 Place the TLC plate under the radiation so that the radiations fall perpendicularly on the plate and examine the spot.
5.2.7 The lamp should be capable of revealing a standard spot of a diameter of about 5 mm of the plate.
5.2.8 The distance between the lamp and the chromatographic plate under examination used in
a pharmacopoeia test should never exceed the distance used to carry out the above test.
5.2.9 Use the calibrated and certified UV light meter for measure the intensity of UV light.
5.2.10 Document the activity of Calibration in Annexure I.
5.2.11 Fill the Calibration Tag and affix it on the instrument.
5.2.12 Report any discrepancy observed during calibration of instrument to Quality Control In-Charge
or his designee for corrective and preventive action.
5.2.13 Quality Control In-Charge or his designee informs the defect to Maintenance Department
and service engineer to rectify the defect. Affix ‘Under Maintenance’ label on the instrument.
5.2.14 Calibration should be required after maintenance.
