1.0 OBJECTIVE

1.1 To lay down a procedure for Operation, cleaning and calibration of Instrument in Quality Control Department

2.0 SCOPE
2.1 This Procedure is applicable for UV & Visible Spectrophotometer.

5.0 PROCEDURE

5.1.1 Switch on the main power supply.
5.1.2 After getting the initialization of instrument About 15 minutes, instrument Triple beep sound.



5.1.3 Then Press enter and again sound of Three beep, Open the software on system UV-Visible
Analyst display on the system screen.
5.1.4 Display System status, close Click on File menu, Click New and select Fixed point
measurement for Single wavelength & multiple wavelength.
5.1.5 Click on File menu, Click New and select Wavelength scan measurement for scanning.
5.1.6 For single or multiple wavelength type No of points then type wavelength, we can use upto 10
wavelength at a time.
5.1.7 For Scan click Wavelength Scan Measurement Fill up the blank medium in both cuvette and click
Z for both cuvette matching.
5.1.8 Then fill sample in sample cuvette and click red arrow.
5.1.9 After completion the test click information and type data such as ample name, Batch number and
Operator.
5.1.10 Click file menu, click save and type name of Batch number of the sample Such as CT-016001
5.1.11 Click file menu, click print.

5.2 Precautions/Cleaning

5.2.1 Ensure that the curettes are clean & dry before use.
5.2.2 After every use of curettes wash with water & then methanol, clean with tissue paper and keep
in curette Box.

5.3 Calibration

5.3.1 CALIBRATION FOR ABSORBANCE

5.3.2 Prepare a solution of potassium dichromate spectroscopy grade as per IP 2014

5.3.3 Take the absorbance of Potassium dichromate solution (60 mg dilute to 1000 ml dissolve in 0.005 M of
H2SO4) at following wavelengths 235 nm, 257 nm ,313 nm, 350 nm
5.3.4 Take the absorbance of Potassium dichromate solution (60 mg dilute to 100 ml dissolve in 0.005 of H2SO4) at 430 nm.
5.3.5 Calculate the (1% 1cm) values corresponding to above wavelengths by following formula.

                                         Absorbance x 10

nm = (235, 257, 313, 350 & 430)

 

Calculated value of E (1% 1cm) should lie between the following permissible

5.3.6 Control of Wavelength
Sample Preparation: Prepare a 4% w/v of Holmium Oxide in 1.4M Perhloric Acid

5.3.8 Prepare 0.02%v/v solution of toluene in hexane spectroscopy grade solvent.

5.3.9 Take the absorbance to the above solution at 269nm and 266nm.

5.3.10 Limit of Stray Limit

5.3.11 Prepare 1.2 % w/v of potassium chloride in water.

5.3.12 Calibration Frequency

Once in 6 months and after each maintenance job.
