1.1 To lay down the Procedure for Operation And Calibration of Micro Pipette of Microbiology Laboratory.

5.1 Requirement
5.1.1 Micro pipette tips, Beaker, Distilled water

5.2.1 Fix the tip on nose cone of the micro pipette
5.2.2 Set the desired volume by volume control knob.
5.2.3 Hold the pipette vertically while filling; press the piston button to the first stop.
5.2.4 Immerse the tip of the pipette 2-3mm into the liquid.
5.2.5 Slowly allow the push button to glide back (never slap back).
5.2.6 Slide the tip out, along the inside of the vessel.
5.2.7 Wipe off any droplet from the outer surface of the tip.
5.2.8 Rinse the tip 2 or 3 times with the liquid to be pipetted.
5.2.9 To empty the tip, press the button down to the first stop and wait for 1 to 3 seconds.
5.2.10Press button to the second stop and blow out to empty the remaining liquid.
5.2.12Calibrate the micropipettes in a vibration free room at a constant temperature of water between 20˚C to 25˚C
5.2.13 Calibrate the micropipette as per given in table I
5.2.14First determine the density factor of water and then set the desired volume using the volume setting knob.
5.2.15Carefully fit the tip in tip cone and flush with distilled water by pipetting the adjusted volume at least 3-4 times.
5.2.16Carefully aspirate the liquid and pipette the sucked volume of distilled water in tarred beaker.
5.2.17Repeat at least ten times and record the result.
5.2.18Calculate the pipette volume as per the formula given below
Volume of Water = Weight of water
Specific gravity of water at 25˚C
5.2.19 The percentage variation can be calculated by formula given below
% Variation = 100 x Actual weight –Observed weight
