1.1 To lay down the Procedure for Monitoring of Compressed Air/gases for microbiological analysis.
2.1 This procedure applicable for Monitoring of Compressed Air/gases( nitrogen, carbon, & oxygen) for microbiological analysis for Microbiology Laboratory

5.1.1 70% IPA
5.2.1 Prepare the Soyabean Casein Digest Medium as per SOP for media preparation.
5.2.2 Dispense 100ml of SCDM if cleaned gas sampling assembly. Close the inlet and outlet tube with air 0.2µvent filter.
5.2.3 Sterilize the assembly as per SOP.
5.2.4 Pre-incubated the assembly with media at 30-35°C for 48hours before use.
5.2.5 Bring the sample assembly to sampling location.
5.2.6 Disinfect the outer surface of sampling point by the spraying of 70%V/V IPA.
5.2.7 Flush the gas sample for 1 min.
5.2.8 Open the air/gas sample valve slowly and take the sample for 10min.
5.2.9 After collecting the sample replace the vent filter of inlet tubing and label the assembly with appropriate details. i.e. Sample identification, Date.
5.2.10 Bring the sample to the laboratory and incubate assembly at 23°C ± 2°C for 7 days followed by incubation at 33°C ± 2°C for 7 days. In case of nitrogen samples, incubate the samples in anaerobic conditions.
5.2.11 After completion the incubation record the results.
