1.0 OBJECTIVE :
The objective of this SOP is:
1.1 To lay down a procedure for swab sampling and testing for clean rooms of manufacturing area.
2.0 RESPONSIBILITY:
Microbiologist QC
3.0 ACCOUNTABILITY:
Head – Quality Control
4.0 PROCEDURE:
4.1 Prepare the settle plates as per SOP
4.2 Carry required number of swabs to the concerned areas by putting into a air tight S-S container.
4.3 Enter in the respective areas by following the proper entry ( gowning ) procedure.
4.4 Follow SOP for transferring sampling material containers in the aseptic area.
4.5 Open the container in the respective areas and take the swab sample ( 30cm2 )from the locations mentioned in annexure with
the help of sterile screw caped sealed swab procured ready made from Hi-Media Laboratories Ltd. Mumbai.
4.6 After collecting swab samples from respective areas carry those swab samples to microbiology laboratory.
4.7 Rinse the swab thoroughly into 100ml sterile peptone solution and wash the 0.22 membrane filter ,wash with 3 x 100ml
sterile peptone water and then inoculate in sterile SCD agar plate
4.8 Incubate those plates, in a invert position at 20-25 C for 72 hrs. Followed by 30-35 C for 48 hrs.
4.9 After completion of the incubation period observe the plates and note down the observations in the format mentioned in Monitoring plan.
4.10 Frequency, limits and locations are mentioned in the monitoring plan.
4.11 Store the records in documentation room.
5.0 REASON FOR REVISION:
Change in Environmental monitoring Plan.
