Disposal of Used Culture and Used Media
1.0 Objective
To lay down a procedure for disposal of used culture and used media.
2.0 Scope
This Standard Operating Procedure is applicable for disposal of used culture and used media to be followed at formulation plants of abc company.
3.0 Responsibility
3.1 Officer / Executive Quality Control Department are responsible for disposal of used culture and used media.
3.2 Head Quality control is responsible for the compliance of this SOP.

5.0 Procedure
5.1 For Microbial Culture:
5.1.1 After each subculture, check the growth of the respective cultures after the recommended incubation period and if the growth is satisfactory, the earlier culture tube shall be disposed off.
5.1.2 After the requisite time period for the preservation of any culture is over, remove the culture tubes from the refrigerator.
5.1.3 Sterilize the culture tubes at 121 °C temperature for 30 Min.
5.1.4 After completion of sterilization cycle, wear hand gloves and remove the culture tubes from the autoclave.
5.1.5 Send autoclaved (decontaminated) content in liquid form to the ETP for disposal.
5.1.6 Remove any labels / markings on tube.
5.1.7 Dip the empty tubes in disinfectant for 30 minutes.
5.1.8 Wash the test tubes thoroughly in the running purified water.
5.2 For Culture Media Plates:
5.2.1 Place the entire Petri plate along with the media contents in a disposable bags or metal container.
5.2.2 Autoclave the container at 121°C for 30 minutes.
5.2.3 The distorted Petri plates are sent to the scrap yard and decontaminated media are sent to the ETP for disposal.
5.2.4 Dip the Petri plates in disinfectant for 30 minutes. Wash the Petri plates thoroughly in the running purified water.
5.3 For expired media:
5.3.1 Wear hand gloves and nose mask.
5.3.2 Remove the contents of the media container with the help of spatula into a bucket having purified water.
5.3.3 Dissolve the media in purified water properly and then transfer the contents in disposable bag or metallic container.
5.3.4 Autoclave the disposable bag/container at 121°C for 30 minutes.
5.3.5 Send autoclave (decontaminated) content to the ETP for disposal.
5.3.6 Maintain the Disposal record of used culture and used media in Used Media sterilization Record.
