Cleaning and Sterilization of Glassware

5.0 PROCEDURE
5.1 Requirement
5.1.1 Stainless Steel trays
5.1.2 Glassware
5.1.3 Disinfectant solution.
5.1.4 Liquid detergent.
5.2 Methodology
5.2.1 Wear a gloves and nose mask, and then collect all the Glassware to be washed in Stainless Steel trays.
5.2.2 Contaminated media containing tubes, Petri plates & flasks, do not open any tube, Petri plate or flask.
First sterilize these contaminated materials at 121ºC & 15 psi for 30 minutes.
5.2.3 Remove the content from the tubes and Petri plates into a poly bag and kept aside for disposal.
5.2.4 After removal the media, first glassware should be soaked in Teepol 0.1% solution for 30 Minutes. And then washed
with Purified water and then add in the IPA solution.
5.2.5 Clean the Glassware with liquid soap solution. Use nylon brush to remove all the stains.
5.2.6 For new Glassware, soak in chromic acid mixture for 2–3 minutes using rubber hand gloves. Rinse with Purified water to
remove the traces of chromic acid and finally clean with liquid soap. Use nylon brush to remove all the stains.
Prepare chromic acid as per IP.
5.2.7 Rinse the glassware with Purified water to remove the soap solution completely as detected physically by absence of any foam.
5.2.8 Allow to drain excess water from the washed Glassware by keeping them in inverted position in a clean crate.
5.2.9 Dry the glassware in Hot Air Oven at 60oC for 1 hour. After the Drying period is over allow the glassware to cool and use it.
5.2.10 After drying, wrap the Glassware and sterilize at 1210C at 15 psi for 30 minutes.
