1.0. OBJECTIVE
1.1 To lay down a procedure for the collection & preservation of the in-house isolated Microorganisms .
2.0. SCOPE :
2.1 This procedure is applicable for the collection & preservation of the in-house isolated Microorganisms in
Microbiology Laboratory of the Quality control department
3.0. RESPONSIBILITY :
3.1 Sr.Officer / Officer – Quality Control
To identify and select the frequently occurring colonies for in-house isolated micro organism .
To Perform the gram staining for identification purpose .
Dy.Manager/Designee – Quality Control
To review the gram staining for identification purpose .
4.0. ACCOUNTABILITY :
4.1 Head – Quality Control Department
4.1.1 Head of the department is accountable to ensure the compliance of SOP for all aspects .
5.0. PROCEDURE :
5.1 Observe the exposed settle plates, air count plates, water analysis plates and swabs of machine
surface of sterile aseptic areas for every 7th day for 2 weeks .
5.2 Try to identify colonies of frequently occurring microorganism by shape, size and colour of
the colonies obtained on the exposed settle plates, air count plates, water analysis plates and swabs of machine
surface of sterile aseptic areas.
5.3 Identify organisms morphologically under microscope by gram staining .
5.4. Select minimum eight such types of colonies and aseptically transfer individually each
colony by a transfer loop on to nutrient agar slant /Soyabean casein digest agar slant for bacterial cultures and
Potato dextrose agar /Sabouraud dextrose agar for fungal cultures .
5.5. Assign name to individually culture slant as BIHC(Bacterial in-house culture) for bacterial in house culture and FIHC (Fungal in-house culture) and assign number to individually culture slant as 01 to 08 .eg. BIHC-01, BIHC-02 …… .
5.6. Send the all isolated in house cultures to approved out side lab for identification up to species level .
5.7 Maintain these cultures as per SOP No.
5.8 If in routine observation any colony observed frequently as per point no. 5.2 ,Isolate that and include as in-house culture as per above mentioned procedure .
5.9 PRESERVATION OF MICROBIAL CULTURES :
5.9.1 Preserve well grown microbial culture slant under refrigeration at 2° C to 8° C .
5.9.2 In case if any contamination is observed discard the slant and proceed for fresh
Sub culturing of the same culture .
