5.2 Methodology
5.3 The dehydrated culture media as well as their ingredients are highly hygroscopic and must be stored in a cool dry place away from bright light.
5.3.1 Carefully read the instructions printed on the label of dehydrated culture media containers. Check the composition of the medium and the direction for its use.
5.3.2 Examine the physical nature of the dehydrated medium. If any unusual color, odor or physical appearance is noticed, discard the medium.
5.3.3 Always use a dry spoon or spatula for weighing the dehydrated media. The weighing operation shall be completed as quickly

as possible to prevent absorption of moisture by the hygroscopic contents. Wear hand gloves & nose mask while weighing the

dehydrated media to avoid inhalation of fine particles of media.
5.3.4 Operate the balance and weigh accurately the required quantity of dehydrated culture medium on a butter paper transfer the powder to a suitable container.
5.3.5 Add a part of the required quantity of Purified Water and mix the contents. Make up the volume with Purified Water & mix well.
5.3.6 Check the pH of media at 25ºC±2ºC. If there is requirement of pH adjustment, adjust the pH with 1M NaOH or 1M HCl.
5.3.7 Now heat the contents with frequent agitation till the contents are homogenous.
5.3.8 Dispense the reconstituted medium in suitable containers like test tubes, bottles or flasks. The volume of these containers shall be sufficiently

large to facilitate proper aeration and avoid overflow of medium during sterilization.
5.3.9 All culture media containers shall be plugged with non absorbent cotton and covered with a butter paper/aluminum foil.
5.3.10 All the media tubes and flasks should be labeled with details like Name of Media, Media Lot No., Date of Preparation.
5.3.11 Media after preparation and dispensing as above shall be sterilized as soon as possible. Media prepared and dispensed as

above shall be sterilized in an autoclave at 121°C/115°C for 15 minutes.
5.3.12 Specified limits as directed on label of media bottle, discard the media as per SOP for destruction of microbial waste and contaminated media by autoclave SOP.
5.3.13 If the medium is not used in the same day of preparation, it should be stored properly for not more than 7 days.
5.3.14 Stored Media showing any change in appearance or any sign of visible microbial contamination or cracks or dehydration shall be discarded.
5.3.15 Enter details of prepared media in the ‘Media Preparation & Consumption Record’
