SOP for Operation & Calibration of pH meter in Micro Department
1.0 OBJECTIVE
To provide the procedure of Operation & Calibration of pH meter in Micro Department.
2.0 SCOPE
This procedure is applicable for Operation & Calibration of pH meter

5.0 PROCEDURE
5.1.1 Connect the plug of the pH meter to the power supply and switch ‘ON ‘the power supply.
5.1.2 Switch ‘ON ‘the pH meter. Before measuring the sample wash the pH electrode rinse with
Purified water and clean properly with tissue paper.
5.1.3 Dip the electrode into sample bottle or other sample container and observe the value from display.
5.2 Cleaning
5.2.1 Before using check that the all external parts of the pH meter are clean.
5.2.2 Cleaning is to be done by using soft lint free duster.
5.2.3 For cleaning 70 % IPA or other approved disinfectant solution is to be use.
5.3 CALIBRATION
5.3.1 The first press the calibration mode.
5.3.2 Immerse pH electrode in buffer solution pH 7.00 and record the reading display on Screen
if reading is not between limits. Calibrate the instrument by Rotating calibration knob.
5.3.3 Remove pH electrode from first buffer and rinse with purified water properly and clean with
tissue paper.
5.3.4 Immerse pH electrode in buffer solution pH 4.01 and record the reading display on Screen
if reading is not between limits. Calibrate the instrument by Rotating calibration knob.
5.3.5 Remove pH electrode from second buffer and rinse with purified water properly and clean with
tissue Paper.
5.3.6 Immerse pH electrode in buffer solution pH 9.20 and record the reading display on Screen if
reading is not between limits. Calibrate the instrument by Rotating calibration knob.
5.3.7 Fill the observe value in (instrument calibration records log book .)and
label the instrument as per log book
5.4 FREQUENCY OF CALIBRATION
Daily and after each maintenance.
5.4.1 ACCEPTANCE CRITERIA
For Buffer pH- 4.2 (±0.5) Buffer pH- 7.0, (±0.5) Buffer pH- 9.20 (±0.5)
