SOP for operation cleaning & calibration of Digital colony counter
1.0 OBJECTIVE
1.1 To lay down the procedure for operation , cleaning & calibration of Digital colony counter.
2.0 SCOPE
2.1 This SOP is applicable for operation, cleaning & calibration of digital colony counter which is installed in microbiology department

5.0 PROCEDURE
5.1 CLEANING
5.1.1 De-dust the external surface of colony counter and magnifier with lint free duster. Wipe the outer surface of the instrument with 70 % IPA solution.
5.2 OPERATIONAL PROCEDURE
5.2.1 Connect the colony counter to the main switch “ON” the power switch.
5.2.2 Now switch “ON” the mains switch of the equipment.
5.2.3 Now the digital display show “0000”, if the digital display is not showing “0000” then press the reset button, digital display will show “0000”.
5.2.4 Place a Petri dish on illuminating surface.
5.2.5 Adjust the magnifying glass to view a clear colony present on the Petri dish.
5.2.6 Open the lid of marker pen and properly press the tip of the pen on the colonies present on the Petri dish and checks the digital
display will show you a colony in number.
5.2.7 Record the colony count in formats and remove the plate from the platform.
5.2.8 Press the reset button and repeat the above procedure for all the plates.
5.2.9 After completion of work press the reset button switch OFF the mains.
5.3 CALIBRATION:
5.3.1 At the time of calibration, operate the instrument as per the operating procedure and press the tip of the marker 10,,20,50,100,150,200 time
and then check the number displayed on the screen. Reset ‘000’ and again press the manual button 10,20,50,100,150,200 times and check
the number display on the screen. In both the cases the number displayed on the screen is also 10,20,50,100,150,200
