1.0 OBJECTIVE
1.1 To lay down a procedure for Receipt, Storage and Usage of Culture Media.

2.1 This procedure applicable for Receipt , Storage and Usage of Culture Media In Microbiology Laboratory

5.0 PROCEDURE
5.1 Receipt
5.1.1 Upon receipt of media in QC, check the media for the shipment condition.
5.1.2 Verify with the requisition, and if it tallies put a tick mark and initialize with the date of receipt.
5.1.3 Identify and circle the expiration date on the manufacturer’s label.
5.1.4 Paste the material receipt label, enter the date of receipt on the label.
5.1.5 Assign in-house expiry date (use before date) of one year from the date of opening.
5.1.6 If the manufacturer expiry date is earlier to the in-house expiry date then assign manufacturers expiry date.
5.1.7 Assign a serial number to each container.
5.1.8 Document the details of media batch no or lot no, date of receipt and the serial number allotted to each container in the inventory record.
5.1.9 If the media has to be dispensed into smaller containers, dispense the media from the bulk container to the test tube and check the pH before dispensing.

5.2 Storage
5.2.1 Store the media as directed on the manufacturer’s label. (i.e. a temperature below 25ºC in a dry place). The media for which the recommended storage temperature.
5.2.2 Protect the media from bright light.
5.3 Usage
5.3.1 Use ‘First In First Out’ system by selecting the lowest serially numbered container.
5.3.2 When a new bottle of the media is taken for use, enter the date of opening in the inventory record against the same serial number as on the container.
5.3.3 Enter the date of opening on the material receipt label.
5.3.4 Check the use before date on the container before opening the bottle.
5.3.5 Check the contents for the free flowing condition, after opening the bottle.
5.3.6 Discard the media that shows signs of hydration, cracking or discoloration.
5.3.7 Document the details in the media weighing record.
5.3.8 Calculate the required quantity of the medium on the basis of manufacturer’s instructions.
5.3.9 Weigh the calculated quantity on a calibrated balance, quickly and accurately.
5.3.10 Immediately close the bottle and tighten the lid.
5.3.11 Document the weighing in the media weighing record.
5.3.12 Add correct volume of WFI and mix thoroughly.
5.3.13 If the medium contains agar, leave the flask to stand, allow the agar particles to hydrate and then heat the media with frequent agitation, to prevent charring. Boil the media till the agar dissolves.
5.3.14 After completion of heating, add 0.5% of glycerol to Soya bean casein digest agar.
5.3.15 Dispense the media into the required containers with constant shaking.
5.3.16 Plug the container and mark every media container with media type, Date of preparation and used before Date.
5.3.17 Mark each media container for the volume level before sterilization.
5.3.18 Dispense the media into a separate screw capped tube (test tube) to check the pH.
5.3.19 Check the pH before sterilization, for liquid media at room temperature, and for agar media at 40-45 °C.
5.3.20 Adjust pH if required by using Sodium hydroxide or Hydrochloric Acid before sterilization.
5.3.21 Autoclave the media in same test tube to check the pH of the medium after sterilization and cooling.
