Operation and Calibration of Air Sampler
1.0 Objective
To lay down a procedure for Operation and calibration of Air sampler.
2.0 Scope
This Standard Operating Procedure at Microbiology laboratory Quality control department of abc company.
3.0 Responsibility
3.1 Concerned Officer/Executive shall be responsible to follow this SOP.
3.2 Department Head or his designee shall be responsible to implement and ensuring to compliance of this SOP.

5.0 Procedure
5.1 Operation and calibration
5.1.1 Connect the power cable to instrument and power supply, Recharge the instrument for an one hour before for every usage.
5.1.2 Switch ON the instrument by once single pressing on the ‘ON/OFF’ button.
5.1.3 Set the time for 1000 liters by using ‘Set’ and ‘Arrow’ buttons.
5.1.4 Carry the sterile SS stain cup and Petri plates in a sterile condition to area where the Air sampling is scheduled.
5.1.5 Open the SS stain cup, and assemble the Petri plate in a direction to open side of air flow.
5.1.6 Prepare the media and Petri plate.
5.1.7 Once the Petri plates are assembled, press the start button.
5.1.8 After 1000 liters completed air vacuum pump will gets stops automatically.
5.1.9 Open the SS stain cup and remove the Petri plate from the instrument, close the Petri plate with upper part of Petri plate immediately.
5.1.10 Carry the Petri plate in a sterile condition to the Microbiology lab.
5.1.11 Keep the plates for incubation as per respective SOP No BBL/QC/SOP/028.
5.2 Cleaning and Sterilization
5.2.1 Before using the air sampler, wrapped SS stain cup in butter paper and keep it in Autoclave for Sterilization.
5.2.2 Sanitize the tubular metallic handle with 70 % v/v Isopropyl alcohol.
5.2.3 Clean the impeller with 70 % v/v Isopropyl alcohol between sampling cycles.
5.3 Precautions
5.3.1 Do not autoclave the tubular stainless body, it will damage the system.
5.3.2 Ensure the battery power is sufficient for conducting the test well in advance.
5.3.3 Test the power fall of the battery by switching on the control box and note the alarm, which should occur at any given timer selection.
5.3.4 For the charging the battery, bring the DC/AC switch on the neutral position and connect to 220V AC mains, using power cord.
5.3.5 After completion of the work put the air sampler into its carry box.
5.3.6 Calibration shall be performed for Motor /Battery efficiency, and for Air flow rate with calibrated anemometer by authorized out source calibration party once a year and when ever a major maintenance is done.
5.3.7 Acceptance criteria: 180 liters air per minute.
